Targeting IgE and Th2-Cytokines in Allergy: Brief Updates on Monoclonal Antibodies and Antibody Gene Therapy

چکیده
The search for an effective treatment of allergic conditions is ongoing global health challenge due to the high prevalence allergies. Epinephrine and glucocorticosteroids remain oldest most widely used regimen allergy, these medications are short relief. In extreme allergy manifestations, current options aim use monoclonal antibody (mAb) target pathological pathways inflammation involving mast cells, eosinophils, basophils. These cells have propensity induce allergic-inflammatory response. Studies shown that they responsible several diseases, such as asthma, atopic dermatitis, rhinitis, conjunctivitis. evaluating antibodies against serum IgE (Omalizumab), Th-2 cytokines, IL-4, IL-13 (dupilumab), IL-5 suggest attenuation symptoms improvement in patients’ overall well-being. However, factors cost production (i.e., purification), host immunogenicity, safety, efficacy hindered availability purified mAb developing countries. Gene therapy a promising tool treating emerging studies suggested gene may be future cases manifestations. This paper describes severe responses associated limitations. It explores prospects modulating episodes.

IgE and IgG antibodies in skin allergy of the horse.
In horses, allergies have been characterized by clinical signs and/or intradermal (i.d.) allergen testing. Our aim was to find the first direct evidence that immunoglobulin E (IgE) mediates equine allergy. In addition, we tested the hypothesis that immediate skin reactions in horses can also be mediated by IgG. Anti-IgE affinity columns were used to purify IgE from serum of one healthy horse an...

We are commenting on the article by Klein et al. on the functional properties of several anti-CD20 monoclonal antibodies (mAbs) in the January/February 2013 issue of mAbs. 1 Rituximab, the first monoclonal antibody approved for cancer therapy, has truly revolutionized the treatment of a variety of CD20 + hema-tological malignancies, 2,3 and also has been studied in a large number of autoimmune ...

SUMMARY Monoclonal Antibodies are identical antibodies and Can be generated in ultimated amounts by countinuos cultures of single antibody- secreting cells. These cell lines are produced by cell fusion of normal lymphocytes to cells of a myeloma tumor cell line, which confers on the antibody producing hybrid cell, immortality and the ability grow as atumor in animals. monoclonal antibodies c...
متن کاملGeneration of human monoclonal allergen-specific IgE and IgG antibodies from synthetic antibody libraries.
BACKGROUND Allergen-specific IgE and IgG antibodies play pivotal roles in the induction and progression of allergic hypersensitivity reactions. Consequently, monoclonal human IgE and IgG4 antibodies with defined specificity for allergens should be useful in allergy research and diagnostic tests. We used combinatorial antibody libraries and subsequent recombinant production to make and assess Ig...
متن کاملMonoclonal Antibodies and Polyclonal Antibodies: A Brief Comparison
It is called an immunoglobulin that is used for neutralization of the foreign pathogens like viruses, bacteria or any foreign bodies invade the blood. When antibodies produced from immunized animals against certain antigen will produce what is called polyclonal antibodies (PAbs) which are mixture of different antibodies produced by many different B cell clones that identify different epitopes (...
